摘要
目的:建立九分喷雾剂中士的宁和马钱子碱的含量测定方法。方法:采用反相高效液相色谱法(RP-HPLC法)。色谱柱为NucleodurC18柱(5μm,4.6mm×250mm);流动相为甲醇-水(65∶35),1000mL溶液中含十二烷基磺酸钠1g、磷酸二氢钾0.4g;检测波长:254nm。结果:士的宁和马钱子碱回归方程分别为Y=15.51+2772.70X(r=0.9995),Y=6.44+1848.65X(r=0.9992);线性范围分别为0.1747~0.8736μg,0.1088~0.5444μg;平均回收率分别为98.92%,99.43%;RSD分别为1.70%,2.58%。结论:RP-HPLC法可作为九分喷雾剂的质量控制方法之一。
Objective:To establish the RP-HPLC method for determination of strychnine and brucine in the preparation of Jiufen Spraying.Methods:RP-HPLC Column was NUCLEODUR C18 (5 μm,4.6 mm×250 mm).Mobile phase was Methanol-Water(65 ∶35)(1 g Sodium dodecyl sulfonate and 0.4 g KH2PO4 in 1 000 mL Water).The detection wavelength was at 254 nm.Results:Strychnine and brucine had a good linearity (r=0.999 5,0.999 2)in the range of 0.174 7~0.873 6 μg and 0.108 8~0.544 4 μg.The mean recovery were 98.92%(RSD=1.70%)and 99.43%(RSD=2.58%).Conclusion:This method is suitable for quality control of this preparation.
