缺血-再灌注大鼠视网膜TPA变化与视网膜水肿的关系

摘要  目的 探讨缺血-再灌注对大鼠视网膜分泌组织型纤溶酶原激活物(TPA)的影响及其与视网膜水肿的关系。方法 采用提高眼压法造成视网膜缺血后,恢复眼压形成血流再灌注。实验分正常对照组、缺血 1h再灌注 1h组、缺血 1h再灌注 2h组、缺血 2h再灌注 1h组和缺血 2h再灌注 2h组。每组各取 10例测试视网膜组织TPA的活性和含水量。结果 缺血-再灌注后,大鼠视网膜组织TPA的活性和含水量随缺血和再灌注时间的延长而升高(P<0 01)。 结论 缺血-再灌注可引起视网膜组织TPA的活性升高和视网膜水肿,是视网膜组织结构和功能损伤的因素之一。 Objective To explore the relationship between the activity change of tissue type plasminogen activator(TPA) of retinas and retinal edema in retinal ischemia and reperfusion rats. Methods The intraocular pressure(IOP) was raised to creat retinal ischemia for 1 hour,and then IOP was lowered to normal to allow retinal reperfusion for 1 hour in 50 60 day SD rats.The retinal TPA activities and the retinal edema in control group and 4 ischemia reperfusion groups were detected and evaluated. Results TPA activity and water content in retinas were increased after ischemia reperfusion ( P <0 01).The TPA activity of control group was 11 32±1 17μmol/(min·mL),and that of 4 ischemia reperfusion groups was 16 28±2 36,19 14±2 49,20 49±2 55 and 24 18±2 74 respectively.The water content was 78 56% in control group,and that of 4 ischemia reperfusion groups was 88 76%,90 97%,91 21% and 93 65% respectively. Conclusion Ischemia and reperfusion of retina can stimulate the rising of activity of TPA and induce retinal edema.The damage of retina may be associated with the increasing of the activity of TPA.
