摘要
目的 :探讨血小板反应素 (TSP)的表达及微血管密度(MVD)改变与非小细胞肺癌 (NSCLC)的关系 .方法 :用SP免疫组织化学技术 ,检测了 5 5例NSCLC癌组织和 30例肺癌旁组织中TSP1和TSP2的表达及MVD的改变 .结果 :NSCLC癌组织中存在活跃的血管生成 ;MVD与NSCLC侵袭转移和临床分期有关 .肺癌组织中TSP1和TSP2阳性表达的图像分析灰度值(15 8 7± 2 1和 14 9 1± 2 9)明显高于 30例肺癌旁组织阳性表达的图像分析灰度值 (14 1 2± 7 2和 14 5 7± 4 7) (P <0 0 5 ) ;TSP1和TSP2阳性表达与NSCLC患者的病情进展和转移明显有关 .TSP1和TSP2表达与MVD成负相关 (r=- 0 96 4 ,-0 876 ,P <0 0 1) ,且TSP1起更重要的作用 .结论 :TSP1和TSP2与NSCLC的血管生成有密切关系 .TSP1和TSP2及MVD是临床估计NSCLC病情进展和侵袭转移的重要标志 .
AIM: To explore the relationship between the expression of thrombospondin (TSP) and angiogenesis in non-small cell lung cancer (NSCLC). METHODS: By using SP immunohistochemistry stainning method, expression of TSP1 and TSP2 and microvessel counting were evaluated in surgically resected specimens from 55 patients with NSCLC and 30 adjacent normal tissues using the anti-TSP1, anti-TSP2 and anti-CD34 monoclonal antibodies, respectively. RESULTS: Active angiogenesis was observed in NSCLC tumor tissues. The microvessel density (MVD) was related to metastasis and the clinical stage of NSCLC. TSP1, TSP2 expressed mainly in the cytoplasm, nuclear and extracellular matrix. The expression of TSP1 and TSP2 in the 55 tumor tissues was significantly higher than that in the 30 adjacent normal tissues(158.70 ± 2.11 versus 141.20±7.20 and 149.10±2.94 versus 145.70±[yn1] 4.74, respectively, P<0.05). The expression of TSP1 and TSP2 in NSCLC patients was related to the clinical stage and metastasis of NSCLC. There was a negative correlation between TSP1, TSP2 expression and MVD (r= -0.964,-0.876, respectively, P<0.05). TSP1 played a more important role. CONCLUSION: It indicates that TSP1 and TSP2 are closely correlated to the angiogenesis of NSCLC. TSP1 and TSP2, together with MVD, may serve as clinical predictors for the metastasis of NSCLC.
