摘要
目的研究甲状腺素对仔鼠脑成纤维母细胞瘤凋亡相关的RNA结合蛋白(neuroblastoma apoptosis-related RNA-binding protein,NAPOR)基因表达的影响。方法以丙基硫氧嘧啶灌胃复制孕鼠甲状腺功能低下模型,采用逆转录-聚合酶链反应检测仔鼠出生后第1、5、10天仔鼠皮层NAPOR的表达,年龄匹配的正常仔鼠为对照组。结果NAPOR在甲状腺功能低下孕鼠所产仔鼠表达均高于正常对照组;在出生后一段时期内,随着鼠龄增长表达降低。结论转录后水平的调节机制可能参与甲状腺素对脑基因表达的影响。
Objective To study the effect of thyroxine on gene expression of neuroblastoma apoptosis-related RNA-binding protein (NAPOR) in fetus brain. Methods 2-N-propyl-6-thiouracil(PTU) was administrated via esophageal intubation to prepare the model of maternal pregnant hypothyroidism. The fetus rats of postnatal 1, 5, and 10 days were decapitated respectively. Reverse transcribed polymerase chain reaction was carried out to detect the expression of NAPOR gene in cortex while the age-matched normal fetus rats as control. Results The cortex experimental group showed higher expression of NAPOR than normal control, tending to decrease the expression with age. Conclusions These data suggests the regulation at the post-transcription level may play a role in the mechanism for thyroxine regulating gene expression in brain.
