大鼠肺鳞癌中DFF45和Bcl-2蛋白的表达及其意义

摘要 目的 :探讨DNA裂解因子 (DFF4 5 )和凋亡抑制基因bcl 2的蛋白产物在大鼠肺鳞癌癌变转化过程中的变化及其关系。方法 :Wistar大鼠 6 0只 ,其中 5 0只用化学致癌物 3 甲基胆蒽 (MCA)及二乙基亚硝胺 (DEN)碘油溶液于左肺叶支气管灌注诱发大鼠肺鳞状细胞癌 ,10只作为对照。应用免疫组织化学SP法检测DFF4 5和bcl 2蛋白在癌变过程中的变化。结果 :对照组大鼠支气管粘膜上皮细胞区、癌前病变区与肺鳞癌区中 ,DFF4 5蛋白的阳性率逐渐升高 ,分别为 2 0 .0 0 % (2 / 10 )、4 3.75 % (7/ 16 )、5 8.82 % (2 0 / 34) ,肺鳞癌区与对照组大鼠支气管粘膜上皮细胞区相比 ,DFF4 5蛋白阳性率的差异有显著性 (P <0 .0 5 ) ;bcl 2蛋白的阳性率分别为 :0 ,31.2 5 % (5 / 16 ) ,4 7.0 6 % (16 / 34) ,对照组大鼠支气管粘膜上皮细胞区分别与癌前病变区、肺鳞癌区比较差异均有显著性 (P <0 .0 5 )。在大鼠肺鳞癌中 ,DFF4 5与bcl 2呈显著正相关 (Pearson列联系数 =0 .5 872 ,P <0 .0 1)。结论 :bcl 2基因的过表达可能抑制了DFF4 5的裂解 ,协同促进了大鼠肺鳞癌的发生发展。 Objective: To investigate the expression of DNA-fragmentation factor 45(DFF45) and apoptosis suppress gene bcl-2 proteins and their relationships in the carcinogenesis of lung squamous cell carcinoma in rats. Methods: 3-methylcholanthrene(MCA) and diethyinitrosamine (DEN) were used to induce lung squamous cell carcinoma by intra-left-lobar-bronchial-instillation in Wistar rats. 50 rats induced lung cancer and 10 control rats instilled with iodized oil were killed in turn at day 15, 30, 60, 90 and 120 after the first instillation. Expression of DFF45 and bcl-2 proteins was detected by immunohistochemistry. Results: DFF45 protein positive rate were 20.00%(2/10), 43.75%(7/16) and 58.82%(20/34) in normal bronchial epithelium cell region, premalignant lesion region and lung cancer region respectively, there was a significant difference of DFF45 protein positive rate between bronchial epithelium cell and lung cancer (P<0.05). Positive rate of bcl-2 protein expression was 0, 31.25%(5/16) and 47.06%(16/34) respectively, in normal bronchial epithelium cell region compared to that in premalignant lesion region and in lung cancer region respectively, and statistical significance were observed (P 1<0.05;P 2<0.05). In rat lung squamous cell carcinoma, there was a positive relationship between DFF45 and bcl-2 (Pearson coefficient=0.5872, P<0.01). Conclusion: Overexpression of bcl-2 gene may block cleavage of DFF45, and synergetically accelerate the developement of lung squamous cell carcinomoma in rats.
