摘要
目的了解周围神经损伤后其远侧端特异性表达的65KD蛋白对鼠胚脊髓运动神经元的影响。方法选用SD大鼠坐骨神经横断后二周的动物模型,自然凝胶系统电泳分析损伤后神经远侧端中的蛋白组分,将65KD蛋白区带胶条与14天鼠胚脊髓运动神经元联合培养。结果损伤后二周的坐骨神经远侧端中特异性地出现表达增强的65KD蛋白;与含65KD蛋白区带胶条联合培养的脊髓运动神经元细胞存活数量多、突起生长旺盛。结论坐骨神经损伤后其远侧端组织中65KD蛋白表达增强,该蛋白具有促进鼠胚脊髓运动神经元存活与突起生长的作用。
Objective To investigate the effect of 65KD protein of peripheral nerve on the spinal motor neurons of embryonic rat by cell culture. Methods Animal models after transection of sciatic nerve two weeks had been employed. Protein composition in the distal stumps had been analysed by native polyacrylam-ide gel electrophoresis. Native-PAGE strip containing 65KD protein for the co-culture with spinal motor neurons of embryonic rat had been performed. Results In the native-PAGE, protein band of 65KD appeared only and expressed highly in the extract from the distal stump after transection 2 week. In co-culture of the 65KD strip with dissociated spinal motor neurons, motor neurons survived and neurites elongated distinctly. Conclusion 65KD protein expressed highly in distal stump after lessioned sciatic nerve and possessed bioac-tivity to survive motor neurons and elongate its neurites clearly.
