人类ABO血型基因型检测的研究

摘要 ABO血型一直采用血清学方法鉴定ABO血型的基因产物,结果可靠、准确,但不能检测出基因型。本文通过ABO血型基因中几个不同位点的差异,设计出特异性引物,采用PCR-SSP技术特异性地扩增A、B和O基因,该方法简单、快速,可鉴定出6种ABO血型基因型AB、AA、AO、BB、BO、OO。本文利用该技术对240名献血者进行了ABO血型的基因型多态性分析,结果p基因频率0.2292,q基因频率0.2125,r基因频率0.5683。 ABO blood group has been identified by serological techniques for a long time . Serological techniques rely on the detection of ABO - associated proteins and are sensitive with very accurate results, But they can t identify ABO genotypes. We have developed ABO genotyping method by taking advantage of ABO DNA sequence differences. This technique uses PCR - SSP to amplify DNA sequences specificity for the A/B or O alleles. This method identifies six possible ABO genotypes including AR,AA,BB,AO,BO and OO. In this report we demonstrate the specificity of the ABO amplification of 240 Chinese.
