摘要
目的 观察CIK细胞的体外增殖能力及其抗肿瘤特性。方法 用淋巴细胞分离液分离外周血单个核细胞 ,加入不同的细胞因子 (IFN、IL 1、IL 2、CD3 mAb) ,诱导生成CIK细胞 ,观察CIK细胞的扩增情况 ,用流式细胞仪检测其表面标志 ,MTT法测定其杀瘤活性。结果 CIK细胞具有较强的扩增能力 ,经过 30天培养 ,总的细胞数可扩增至 70多倍 ;CIK细胞是一群异质细胞群 ,CD3 + CD56+ 细胞为其主要的效应细胞 ,经过 30天培养显著增加 ,为 (39 90± 8 6 3) % ,其绝对值可增加 30 0 0多倍 ;CIK细胞具有较强的杀瘤能力 ,在第 10天时杀瘤活性为6 0 6 9% ,显著高于LAK、CD3 AK细胞 ,在第 2 0天时达到最高值 ,为 78 92 %。结论 CIK细胞具有较强的扩增和杀瘤能力 ,可为过继免疫治疗提供新的手段。
Objection To observe the proliferation ability in vitro and anti-tumor character of CIK cells.Methods Mononuclear cells were isolated from peripheral blood with ficoll solution.CIK cells from PBMC were induced with different cytokines(IFN-γ、IL-1、IL-2、CD3mAb),the proliferation ability of CIK cells was measured by calculation,the phenotype of CIK cells was analyzed by flow cytometer and the killing tumor cell activity of CIK cells was tested with MTT method.Results CIK cells belong to a heterogeneous group of effector cells possessing strong proliferate ability with over 70-flod expansion after 30 days culture,CD 3 +CD 56 + cells was the major effect cell in CIK cell group had a significant increase in both absolute number and the percentage rate.The absolute unmber of CD 3 +CD 56 + cells reached over 3 000 times and the percentage was 39.90% after 30 days culture.CIK cell possessed significantly higher ability to kill tumor cells than LAK cell and CD3AK cell.It got 60.69% of killing tumor cells activity after 10 days cultured and hit the peak of 78.92% after 20 days.Conclusion CIK cell possessed a strong proliferation ability and anti-tumor effect.It might provide a new method of the immunotherapy to cancer.
