摘要 目的:探讨白细胞介素10(IL-10)基因治疗对非肥胖糖尿病(nonobesediabetic,NOD)鼠1型糖尿病发病率、胰岛炎的影响。方法:4周龄雌性NOD小鼠尾静脉快速注射IL-10表达质粒,注射Ringes液作为对照组。酶联免疫吸附实验(ELISA)检测血清中IL-10的表达,10周龄后每周测定血糖,25周龄HE染色观察未发病的NOD鼠胰岛炎程度。结果:处理组IL10的表达持续14d左右,糖尿病发病率为25%,明显低于对照组(71%)(χ2=5.57,P<0.05),且处理组中未发糖尿病的NOD鼠胰岛炎的严重程度亦低于对照组。结论:IL-10基因给药可以降低NOD鼠1型糖尿病发病率及胰岛炎严重程度。 AIM:To study the effects of interleukin-10(IL-10) gene therapy on type 1 diabetes and insulitis in nonobese diabetic(NOD) mice. METHODS:Four-week-old NOD mice were treated with IL-10 expression plasmid(treatment group) or Ringers(control group) by rapid tail caudal vein injection.Serum IL-10 was detected by enzyme-linked immunosorbent assay(ELISA);blood glucose was measured weekly from 10 weeks old.At the age of 25 weeks,degree of insulitis of the NOD mice in two groups that were not diabetic was studied. RESULTS:Serum IL-10 was detectable for 14 days after injection in the treatment group.The incidence of diabetes in the treatment group was 25%,which was significantly less than that in the control group(71%)(χ2=5.57,P< 0.05),and the degree of insulitis in the treatment group was also less than that in the control group. CONCLUSION:Administration of IL-10 gene can decrease the incidence of type 1 diabetes and lower the degree of insulitis in NOD mice.
