糖尿病大鼠胃平滑肌细胞凋亡及相关基因蛋白表达的意义 被引量:5

Apoptosis of gastric smooth muscle cell and associated gene expression in diabetic rats

摘要 目的:探讨糖尿病大鼠胃平滑肌细胞凋亡以及凋亡相关基因蛋白表达变化,为糖尿病胃轻瘫的发生研究提供理论基础。方法:Wistar大鼠70只,随机分为糖尿病组(链脲菌素STZ60mg/Kg,腹腔注射),对照组(腹腔注射等量的生理盐水)。2个月后大鼠色素胃排空和肠道传输速度测定,TUNEL法原位检测凋亡细胞,免疫组织化学方法和Westernblotting免疫印迹检测bcl-2/bax基因表达。结果:糖尿病组的胃内色素残留率显著增加,小肠传输速率减慢(t=4.26,4.18,P<0.01);糖尿病组平滑肌细胞凋亡细胞指数犤(9.63±2.47)%犦较正常对照组犤(3.23±1.28)%犦明显增高(t=4.02,P<0.01);bcl-2基因表达下调,bax基因表达上调。结论:糖尿病胃肠道动力改变与胃平滑肌细胞凋亡密切相关,bcl-2凋亡途径参与了胃轻瘫的发生。 AIM: To investigate the apoptosis of gastric smooth muscle cells(SMCs) and the changes of its associated gene expression in diabetic rats, so as to provide theoretical evidence for the study of pathogenesis of diabetic gastroparesis. METHODS: Seventy Wistar rats were randomly divided into diabetic group and control group.Rats in the diabetic and control group received intraperitoneal injection of streptozotocin(STZ) 60 mg/kg and saline of the same volume respectively.The rates of pigmen gastric emptying and intestinal conduction velocity were measured after 2 months. Apoptosis cells were detected in situ with the technique of terminal deoxynucleotidyl transferase mediated d-UTP nick end labeling (TUNEL).The gene expression of bcl-2/bax were examined by immunohistochemistry and Western-blotting. RESULTS: The remaining rate of pigmen gastric emptying was significantly increased and intestinal conduction velocity was delayed in the diabetic group (t=4.26, 4.18, P< 0.01). The apoptosis index(AI) of SMCs in the diabetic group[(9.63±2.47)%] was significantly increased as compared with that in the normal control group[(3.23±1.28)%].The gene expression of bcl-2 was down-regulated while that of bax was up-regulated in the diabetic group. CONCLUSION:Diabetic gastrointestinal motility dysfunction is closely associated with the apoptosis of SMCs, and bcl-2 mediated apoptotic pathway plays a role in the occurrence of diabetic gastroparesis.
