摘要
目的:探讨羊睾丸提取液对重金属致损睾丸支持细胞的修复与保护作用,以期为干预男性生育功能障碍提供新思路。方法:选用健康昆明种雄性小白鼠30只,随机将其分为对照组、模型组、给药组,每组小鼠10只。采用醋酸铅制备睾丸受损模型,还原型烟酰胺腺嘌呤二核苷酸磷酸-黄递酶(NADPH-d)组织化学方法结合显微图像分析,观察各组小鼠睾丸支持细胞一氧化氮合酶(nitricoxidesyn-thase,NOS)活性。结果:模型组睾丸支持细胞肿胀变性,模型组NOS活性(A值:0.146±0.023)明显低于对照组(0.298±0.031);给药组支持细胞形态接近对照组,给药组NOS活性(0.236±0.020)明显高于模型组(P<0.01)。对照组体质量明显高于模型组(P<0.01);给药组体质量明显高于模型组(P<0.05);对照组与给药组无明显差异(P>0.05)。对照组睾丸质量明显高于模型组(P<0.05);给药组睾丸质量明显高于模型组(P<0.05)。结论:羊睾丸提取液对铅等重金属所致的睾丸损伤有一定的修复和保护作用。
AIM:To study the repairing and protective effects of goat testis extract on sertoli cells with testis lesion caused by heavy metal. METHODS:Thirty healthy Kunming mice were randomly divided into three groups:control group,model group,treatment group,and 10 rats for each group.The animal model with testis lesion was made by lead acetate.By means of NADPH-diaphorase(NADPH-d) histochemical staining and microimage pattern analysis,nitric oxide synthase(NOS)activity in sertoli cells was observed. RESULTS:Compared with the control group,sertoli cells in the model group were swelling and degenerative,and NOS activity in model group(A:0.146±0.023)was evidently lower than that in control group(0.298 ±0.031).Compared with the model group,the pattern of sertoli cells in the treated group was similar to the control group,NOS activity in treated group(0.236±0.020)was higher than that in model group(P< 0.01).The body mass index in control group was significant higher than that in model group(P< 0.01).The body mass index in treatment group was significantly higher than that in model group(P< 0.05).There was no significantly different between control group and treatment group(P >0.05).The testis mass in control group was significant higher than that in model group(P< 0.05).The testis mass in treatment group was significant higher than that in model group(P< 0.05). CONCLUSION:The goat testis extract has a certain repairing and protective effect on testis lesion caused by heavy metal,such as lead,etc.
