小鼠脑发育中CDK5 mRNA表达变化的研究 被引量:3

摘要 目的 观察周期素依赖性蛋白激酶 5 (CDK5 )mRNA在脑发育中的表达与分布。方法 采用胚胎期至老年期小鼠全胚胎及脑切片的原位杂交染色。结果 ①全胚胎原位杂交染色发现脑内CDK5mRNA从胚胎 10 5d(E10 .5 )开始表达 ,主要分布于神经上皮的套层细胞内 ;②脑切片的原位杂交染色观察到从E14到成年期脑内均见CDK5mRNA阳性信号 ,主要定位于神经元的胞浆与核周 ,于新生期前后表达达高峰 ;分布于包括大脑皮层、海马、丘脑、下丘脑、小脑及许多神经核团。在老年期上述区域CDK5表达均有减弱 ,部分区域呈阴性。结论 表明脑内CDK5作用贯穿了整个脑的发育阶段 ,且主要参与了神经系统的早期发育 ;成年后CDK5可能继续保持对某些区域中神经元的功能调节。 Objective To investigate the expression and distribution of cyclin-dependent kinase 5 (CDK5) mRNA in the developmental brain in mice. Methods Embryonic to aged stage murine embryo and brain sections were stained by whole-mount in situ hybridization technique. Results The expression of CDK5 specific mRNA, distributed mainly in the neuroepithelial mantle layer, was detected in the brain tissue from E10.5 d by whole mount in situ hybridization. Higher and stable levels of CDK5 mRNA were detected from E14 d to adulthood mice. The cellular location of the CDK5 mRNA was confined to both nucleus and cytoplasm. The peak of CDK5 expression was around the prenatal and postnatal periods. The main distribution regions of CDK5 mRNA included the cerebral cortex, hippocampus, thalamus, hypothalamus, cerebellum, and several neural nucleus masses. In the aged brains, the expression of CDK5 mRNA decreased, and some regions were negative or weakly stained. Conclusion The results indicate that the changes of CDK5 expression are parallel to the development of nervous system. CDK5 may be involved in the whole neurodevelopmental stages of brain, especially in the early development of nervous system. CDK5 expression in the adult may be closely associated with the regulatory function of neurons in some regions.
