摘要
目的:探讨研究血管紧张素原(AGT)基因3'端GT重复序列多态性在不同种族群体中的分布特点. 方法:应用PCR扩增、毛细管电泳和Genetic Profiler 自动分析软件进行微卫星基因组扫描分析技术,检测170名新疆哈萨克民族AGT基因的3'端GT重复序列多态性. 结果:新疆哈萨克民族AGT基因3'端GT重复序列存在13种等位基因,按片段由小至大分别命名为A1~A13(GT的重复次数依次为13~25), 扩增长度为160~184 bp, 其频率为0.003~0.197.以扩增片段长度为170 bp的A6等位基因最为常见,其次为A3和A4,分布符合Hardy-Weinberg遗传平衡定律.该位点杂合度为0.862,多态信息量为0.847.结论:新疆哈萨克族人AGT基因3'端GT重复序列等位基因型与其他人种、民族人群不同.
Objective:To study the distribution characteristic of GT repeat sequence of angiotenosigen (AGT) gene in various races. Methods: The GT-repeat polymorphism of AGT gene was identified by polymerase chain reaction (PCR) with fluoreascence labeled primers, capillary electrophoretic and automatic analysis Genetic Profiler for micropreparation Genescan in 170 Kazak of Xinjiang. Results: There were 13 alleles ranging from 160 bp to 184 bp in all subjects. The allele frequencies were 0.003 to 0.197, and allele A6 was more frequent in Kazak. The heterozygosity of GT repeat locus was 0.862, the polymorphic information content (PIC) was 0.847. Allele frequencies of the 2 microsatellite DNA in Kazakans are in good agreement with Hardy-Weinberg equilibrium. Conclusion: The results suggest that GT repeat sequence polymorphisms of angiotensinogen gene in Kazak of Xinjiang are different from various races.
