摘要
目的 :探讨实验用小型猪腮腺细胞在体外原代培养条件及观察其生物学特征。方法 :摘取 4月龄实验用小型猪的腮腺组织 ,应用混合胶原酶多步消化、分离腮腺细胞 ,在含有 5 0ml/L胎牛血清、表皮生长因子、转铁蛋白等的HamF12培养基中培养。通过光镜、电镜、免疫组织化学方法鉴定所得细胞 ,并对其蛋白分泌与调节功能进行初步研究。结果 :首次在体外成功培养小型猪腮腺细胞 ,细胞在体外存活 3周以上。在培养液中加入 5 0ml/L胎牛血清最适于腮腺细胞的生长。培养液中加入异丙肾上腺素能促进培养腮腺细胞的蛋白合成与分泌。结论 :成功建立了小型猪腮腺细胞体外培养方法。体外培养的小型猪腮腺细胞具有蛋白合成及分泌功能。
Objective: To investigate the culture conditions of mini -p ig`s parotid gland cells in vitro and to observe the biological features of the cultured cells.Methods: The parotid gland tissues were obta ined from Chinese mini-pigs aged four months. Isolated cells were obtained by digestion of the parotid gland samples with mixed collagenase and cultured in Ha m F12 medium at 37 ℃ in a humidified and 50 ml/L CO 2 incubator. The characte ristics of the cells were studied by morphological and immunohistochemical obser vation. Results: Epidermoid cells growing in monolayer were obta ined. The cells were positve for amylase and negative for vimentin.Abundant secr etory granules in the cells were observed. The addition of secretagogue to the m edium was the most critical factor for long-term survival to mini-pig parotid gland cells. Ham F12 medium supplemented with φ=5% FBS was optimal for the grow th of cultured mini-pig parotid gland cells.Conclusion: Parotid gland cells cultured from miniature pigs in vitro are establixhed. The cult ured cells have the functions of synthesis and secretion of proteins.
