摘要
目的 :探讨锗 Ge M1 0对黑色素瘤细胞增殖的作用 ,及其对高转移性黑色素瘤细胞与内皮细胞粘连的影响。方法 :用 3H- Td R掺入实验检测了 Ge M1 0对 A375黑色素瘤细胞增殖的影响 ;用细胞粘连抑制实验测定了 Ge M1 0对内皮细胞和 A375黑色素瘤细粘连的影响。结果 :Ge M1 0能抑制 A375黑色素瘤细胞 DNA合成 ,浓度为 1 80~ 30 0 μg/ml时抑制率最大 ,可达 73% ,作用时间 2 4 h各浓度 Ge M1 0抑制效果最强。Ge M1 0能部分阻断由L PS和 TNF刺激的内皮细胞和 A375的粘连 ,和对照组比较有显著性差异。结论 :Ge M1 0抗癌活性不仅表现在对肿瘤细胞的直接抑制增殖作用 ,而且可以抑制高转移性黑色素瘤细胞与内皮细胞粘连 。
Objective: To investigate the anti-tumor effects of GeM10 on the inhibiting tumor growth and blocking tumor-endothelium adhesion. Methods:Anti-tumor cytostatics by 3H-TdR in incorporation test and blocking test of adhesion between endothelial and A375 melanoma cell line were studied. Results :GeM10 could significantly inhibit the 3H-TdR incorporation of A375 melanoma cells, over 70% inhibitory rate could be achieved at the concentration ranging from 180μg/ml to 300μg/ml GeM10, the optional inhibitory dose was 180μg/ml GeM10, and the peak inhibitory rate was at 24 hours. GeM10 could partly block the adhesion between endothelium and A357 melanoma (P<0.05), which might explain why GeM10 could prevent tumor metastasis in the animal model. Conclusion :The GeM10 could not only directly inhibit the tumor growth but prevent tumor metastasis as well. These findings further indicate that the GeM10 may have promise as an anti-tumor drug for clinical use.
