大黄素对肝癌细胞SMMC-7721抑制作用及P_(53)、C-myc蛋白的表达 被引量:14

摘要 目的 研究大黄素对肝癌细胞 SMMC-7721 的生长抑制作用和 P53、C-myc 蛋白的表达。方法 应用 MTT法观察大黄素、5-FU 对 SMMC-7721 的生长抑制作用,免疫组化法染色(SP 法)测定细胞凋亡时 P53、C-myc 蛋白的表达情况。结果 大黄素对肝癌细胞 SMMC-7721 有明显的生长抑制作用,其 IC50为 21.6 mol/L。大黄素对 SMMC-7721的抑制作用优于 5-FU(P<0.05);大黄素作用于细胞后,P53、C-myc 蛋白均有一定程度下调,而 C-myc 下调较 P53明显。结论 大黄素对 SMMC-7721 有明显的生长抑制作用,作用于 SMMC-7721 后 P53、C-myc 蛋白的表达均有下调。 Objective To study the effects of emodin on hepatocelluler cancer cells in growthinhibiting and expression of P53 and C-myc protein. Methods The growthinhibiting of hepatocelluler cancer cells in vitro were measured by MTT assy. The expression of P53 and C-myc protein in apoptosis were checked by immumnohistochemical staining. Results Emodin had remarkable effect of growthinhibiting on hepatocelluler cancer cells. IC50 of emodin SMMC-7721 were 21.65 mol/L. The effects of growthinhibiting on hepatocelluler cancer cells by emodin is better than 5-FU. Conclusions Emodin have obvious effects of growthinhibiting on hepatoccelluler cancer cells. P53 and C-myc protein have both fall after hepatocelluler cancer cells expose to emodin.
