摘要
目的评价骨髓间充质干细胞(BMSCs)移植对大鼠脑缺血后肌力、平衡力及空间学习能力的改善作用。方法栓塞大鼠大脑中动脉,制作40只脑缺血2 h再灌注动物模型,再灌注24 h后对移植组(20只)移植异体BMSCs。采用抓绳实验、平衡木行走实验及水迷宫实验,在缺血再灌注后1周、2周、4周和12周时对大鼠肌力、平衡力及空间学习能力进行检测,观察BMSCs移植后脑缺血大鼠神经功能恢复情况。结果脑缺血可导致大鼠肌力、平衡力及空间学习能力下降。缺血后随再灌注时间延长,移植组与对照组大鼠肌力、平衡力及空间学习能力均有一定程度的恢复,而移植组较对照组有明显改善。结论BMSCs移植可较显著地促进脑缺血大鼠肌力、平衡力及空间学习能力的改善。
Objective To determine whether transplantation of bone marrow stromal cells improves the outcome of muscle strength, abilities of equilibrium and spatial learning after focal ischemia in rats. Methods Forty ischemia-reperfusion rat models were established by transient occlusion of the right middle cerebral artery (MCA) to induce focal cerebral ischemia for 2h, followed by reperfusion. Bone marrow stromal cells were administrated at 24h after reperfusion in the transplantation group. The prehensile traction test, beam-traversing task and Morris water maze were used to evaluate the muscle strength, abilities of equilibrium and spatial learning in rats at 1, 2, 4, 12 weeks respectively post reperfusion. Results Cerebral ischemia led to decreasing of muscle strength, abilities of equilibrium and spatial learning. The muscle strength, abilities of equilibrium and spatial learning of rats in both transplantation and control groups unexceptionally got partly improved along with the time elapsed, but with more satisfactory outcome in transplantation group than in control group. Conclusion These results suggest that bone marrow stromal cell transplantation can improve functional outcome of muscle strength, abilities of equilibrium and spatial learning in rats with cerebral ischemia.
