摘要
目的观察外源Smad7基因对L0 2肝细胞株增殖活性的影响。方法分别以重组腺病毒AdSmad7和对照病毒AdGFP在体外感染L0 2肝细胞 ,以RT PCR和Western blot检测外源Smad7基因表达情况 ,MTT法检测L0 2肝细胞增殖活性 ,流式细胞仪检测其细胞周期变化。结果AdSmad7体外感染L0 2肝细胞后 ,Smad7mRNA和蛋白水平均显著上调。转化生长因子 β(TGFβ)可抑制L0 2细胞增殖 ,诱导其凋亡。导入外源Smad7可拮抗TGFβ的作用。结论利用AdSmad7导入外源Smad7基因可间接促进肝细胞增殖 。
Objective To observe the influence of Smad7 overexpression on cell proliferation of the hepatic cell line L02. Methods The hepatic cell line L02 was infected by AdSmad7 and AdGFP respectively. The expression of Smad7 was determined by RT-PCR and western blot. The cell proliferation was measured by MTT and the distribution of the cell cycle was analyzed by flow cytometry. Results When L02 cells were infected by AdSmad7,the increased expression of Smad7 in hepatocytes was detected by both RT-PCR and western blot. TGFβ could inhibit the proliferation of the hepatic cell line L02 and induce its apoptosis.Smad7 gene delivery into L02 can block the effect of TGFβ. Conclusion Smad7 gene delivery into L02 by AdSmad7 could stimulate hepatocytic proliferation and inhibit its apoptosis.
