摘要
为从分子水平探讨硫酸化多糖与小分子相互作用机理,采用氯磺酸 吡啶法(Wolfrom法)对硫酸软骨 素(CS)进行结构修饰,应用光谱法研究了亚甲蓝(MB)与CS空间定向相互作用的机理,考察了硫取代度对CS抗 凝血活性及MB与CS相互作用的影响.测得MB与不同硫取代度的CS最大结合数分别为73、89、103和119, CS的抗凝血活性随硫取代度的增加而增加.认为MB与CS相互作用具有空间位阻效应,CS的抗凝血功能与磺 酸基有关.
In order to know the mechanism of the interaction between sulfate polysaccharides and small molecules under the molecular level, structural modification of Chondroitin Sulfate (CS) by Wolfrom is adopted. The mechanism of the spatial directional of interaction of Chondroitin Sulfate and Methylene Blue (MB) is studied by absorption spectra, and the influence of the sulfur substitute degree on the interaction and Interference Unit of CS is investigated. The maximum biding numbers of Methylene Blue with different CS samples have been obtained, which are 73, 89. 103 and 119 respectively. Interference Unit of CS increases as the sulfur substitute degree increases. It is considered that the interaction between Methylene Blue and Chondroitin Sulfate has an effect on spatial locality interference and Interference Unit of CS is related to its sulfonic group.
