摘要
目的 研究大鼠脑胶质瘤伽玛刀治疗的机制 ,初步探讨胶质瘤细胞凋亡和坏死的发生规律。方法 立体定向法制作C6大鼠脑胶质瘤模型 18只 ,分为实验一、二组和对照组 ,实验组于肿瘤接种后 2 1d行伽玛刀治疗 ,实验一组、实验二组照射剂量分别为 9Gy、12Gy ,治疗后 1周行病理检查及流式细胞学检查测定肿瘤细胞的凋亡和坏死 ,分析凋亡率和坏死率与照射剂量的关系。结果 实验组坏死特点明显不同于对照组 ,实验组可见肿瘤细胞凋亡。流式细胞学检查实验一、二组肿瘤细胞的凋亡和坏死明显高于对照组 ,随着照射剂量的增加 ,肿瘤的坏死增加、凋亡反而降低。结论 肿瘤细胞的凋亡和坏死皆为伽玛刀治疗胶质瘤的作用机制 ,提高肿瘤细胞的凋亡具有重要临床意义。
Objective To study the glioma cells apoptosis and necrosis after gamma-knife radiotherapy in C6 glioma rat.Methods 18 C6 glioma rat model were established by the way of stereotactic injection,and divided into control group and two treatment groups.Two treatment groups were treated by gamma knife irradiation 9Gy、12Gy respectively.To observe the apoptosis and necrosis by flow cytometry.Results Apoptosis(5.392±1.573、3.480±1.727) and necrosis(25.055±6.619、37.230±5.210) were increased significantly more than control group(apoptosis 0.994±0.736、necrosis 8.106±7.895) in both two treatment groups after gamma knife irradiation.However,necrosis was increased along with more irradiation dose administrated while apoptosis was decreased.Conclusion Both apoptosis and necrosis are present after gamma knife irradiation.It is important to increase apoptosis as can as possible for clinic glioma treatment.
