摘要
目的 检测基质金属蛋白酶 9及其抑制物 1的mRNA(MMP9mRNA、TIMP1mRNA)在先兆子痫患者胎盘中的表达 ,以探求MMP9在先兆子痫发病机制中的意义。方法 用半定量逆转录聚合酶链反应 (RT -PCR)技术检测正常妊娠(10例 ) ,先兆子痫 (2 2例 )剖宫产后胎盘中MMP9、TIMP1基因在转录水平的表达。结果 重度先兆子痫胎盘中MMP9mRNA表达量降低 (0 .0 5± 0 .0 4 ) ,TIMP1mRNA表达量增加 ,MMP9/TIMP1比值下降 (0 .0 4± 0 .0 3) ,与轻度先兆子痫及正常对照组比较有明显意义。结论 MMP9表达在基因转录水平就已经下调 ,同时因TIMP1表达增强其活性进一步被抑制 ,因而在先兆子痫的发生和发展中有重要意义。
Objective: quantitatively determining MMP9mRNA、TIMP1mRNA in the placenta tissue by reverse transcription -polymerase chain reaction (RT-PCR) in order to evaluate the role of MMP9 in preeclamosia. Methods: Study group was consisted of 22 women with preeclampsia (severe 14,slight 8),and the control group was consisted of 10 normal pregnancies. The expression of MMP9mRNA、TIMP1mRNA in the placenta tissue was detected by RT-PCR,and their quantities were noted by the density of MMP9/β-actin or TIMP1/β-actin. Results: The expression level of MMP9 of severe preeclampsia group was significant lower (0.05±0.04)than the control group and slight preeclampsia group. And the ratio of MMP9 and TIMP1 was significant lower(0.04±0.03). Conclusion:The results suggested that the expression of MMP9 was already reduced at the level of transcription, and increased expression of TIMP1 mRNA inhibited its activity furthmore.MMP9 has the importance in the forming and development of preeclamosia.
