摘要
目的观察烫伤大鼠小肠肌间神经丛bax/bcl 2蛋白的表达特征与烫伤性细胞凋亡的关系。方法动物分成对照组和烫伤组 ,烫伤组又分 2 4、4 8、72h等几个不同时间点 ,每组于不同时间点进行麻醉、灌注、取材、后固定 ,制成肠肌间神经丛铺片标本。采用免疫组织化学方法 ,进行bax/bcl 2检测。结果bax/bcl 2的表达在烫伤后 2 4h阳性最强 ,以后随着时间延长而逐渐下降。结论细胞凋亡是大鼠严重烫伤后小肠肌间神经丛神经元丢失的重要原因 ,而bax/bcl 2是参与神经细胞凋亡的重要凋控基因。推测烫伤激活bcl 2蛋白的表达是体内最重要的抗凋亡机制之一。
Objective To study the relationship between t he expression of bcl-2/bax protein in the intestinal Auerboch plexus an d apoptosis after the burn in rat. Methods Healthy SD rats were divided into two groups at ran dom:①Contrast group, ②burned group.The burned group was redivided into three groups,the animals of which were operated respectively at 24, 48 and 72 hours after burn and the rats in each groups were anaesthetized, perfusion-fixed int estine was taken out and further fixed for 4 hour(4℃).The outer longitudinal t unica muscularis was peeled carefully from the ring tunica muscularis and tela s ubmucosa ,then the specimens were made. Immunohistochemistry staiming (sp method )was used to detect Bax/bcl-2 protein expression in the intestinal Auerboch pl exus.Results A significant increase in Bax/bcl-2 positive exp ression was detected at 24 hours ,began to decrease at 48 hours and showed much lower at 72 hours post-burn. Conclusion The apoptosis is main cause of the neuronal loss in the intestinal Auerbach plexus in rats after seve re burn.The Bax-bcl-2 is significant regulation gene which praticipate in the apoptosis. It was suggested that the bcl-2 geneexpression in the intes tinal Auerbach plexus after the rat burn-reperfusion insult play an impo rtant role in regulating apoptosis.
