摘要
应用聚丙烯酰胺梯度浓度凝胶电泳法对稻瘟病菌两性菌株的菌体可溶性蛋白质和酯酶同工酶进行了分析比较。结果表明:相同培养条件下的两性菌株的菌体酯酶酶谱相对稳定,同工酶谱带相应分布在凝胶浓度14~26%范围内;交配型A的酯酶谱带明显区别于交配型a,两者相似系数为0.762~0.857;电泳结果,两性菌株的菌体可溶性蛋白质谱带有22~25条,分布在凝胶浓度5~26%范围内;研究结果认为酯酶酶谱或可溶性蛋白质图谱可以作为初步、快速鉴别两性菌株交配型的有效手段。此外,本文还初步探讨了酶谱分析技术在稻瘟病菌分类和演化研究上的意义。
The easterase isoenzymes and soluble proteins of normal hermaphroditic isolates in Pyricu-laria oryzae Cav. were analyzed by CC+PAGE (gradient concentration polyacrylamide gel elec-trophoresis). The results showed that easterase zymograms were relatively stable and isoenzyme nands ranged gradually from 4% to 26% gel in same cultural condition. The zymograms of Mating type A were quite different from Mating type a, and SC (similar cofficient) was 0.762-0.857. The numbers of soluble protein band were between 22-25, ranging from 5% to 27% in gel concentration. The diversity of those gel scanning patterens was obvious. It is suggested that easterase zymogram or soluble protein profile analysis is aeffective method to discriminate mating type of herma-phroditic isolate of P. oryzace Cav. Apart from above, the significance of isoenzyme analysis technique on evolution and classification of P. oryzae Cav. is discussed in this paper.
