20(R)-人参皂甙Rg3对K562/ADM细胞凋亡诱导的研究 被引量:30

摘要 目的 :探讨抗癌药物 2 0 (R) -人参皂甙Rg3(2 0 (R) -GinsenosideRg3)对肿瘤细胞凋亡的诱导。方法 :采用人红白血病耐阿霉素 (adriamycin ,ADM)细胞株K5 6 2 /ADM细胞作为实验模型 ,用ADM作阳性对照。结果 :MTT法细胞毒实验显示 ,Rg3对K5 6 2 /ADM细胞的IC 5 0为 10 4 9± 0 30 μg/ml;流式细胞仪法测定该细胞与浓度为 1 0 μg/ml(无毒剂量 )和 5 0 μg/ml(低毒剂量 )的Rg3共同培养 4 8h的细胞凋亡率分别为 3 39± 0 2 5 %和 9 36± 0 6 1% ;电镜下 ,可见凋亡的K5 6 2 /ADM细胞及凋亡小体。结论 :Rg3具有较强的诱导肿瘤细胞凋亡的能力 ,其诱导强度与其浓度呈正相关 ,对作用时间有依赖性。 Objective:To investigate apoptosis of tumor cells induced by 20(R)-Ginsenoside Rg3.Methods: The human erythroleukemia variant cell K562/ADM that has resistance to adriamycin(ADM) was used an experimental model.The toxicity of Rg3 to K562/ADM cell was tested by method MTT; The apoptosis rate was detected with flow cytometry(FCM); Transmission electron microscope (TEM) was used to find out Apoptotic bodies. Results: The IC50 value of Rg3 on K562/ADM cell was 10 49±0 30 μg/ml;The apoptosis rates of K562/ADM cells with Rg3 1 0 μg/ml(innocurity) and 5 0 μg/ml(low toxicity) were 3 39±0 25% and 9 36±0 61% respectively after cultured for 48 h; Apoptotic bodies of K562/ADM cell were seen by TEM.Conclusions: Rg3 has more powerful ability to induce tumor cell to apotosis; The strength of apoptosis induction of Rg3 for K562/ADM cells is positively relative to the concentrations of Rg3 and has dependence on time.
