摘要
目的 :探讨VEGF促血管生成能力及与肉瘤恶性增殖的相关性。方法 :采用免疫组织化学SABC法检测 6 2例不同组织学分级肉瘤中VEGF、CD3 4 、PCNA基因产物表达情况。结果 :VEGF在Ⅰ、Ⅱ、Ⅲ级肉瘤中的阳性表达率分别 4 1 2 % (7/ 17)、89 3% (2 5 / 2 8)、94 % (16 / 17) ,计数CD3 4 抗体染色阳性的血管内皮细胞 ,算得MVD最低为 3,最高为 113,平均为 32 6 ,肉瘤中PCNA的表达强度分布为 :“ -”5例 (8% )、“ +”10例 (16 1% )、“ + +”31例 (5 0 % )、“ + + +”16例 (2 5 8% )。VEGF与肉瘤的临床病理分级、MVD及PCNA的表达呈显著相关 (P <0 0 5 )。结论 :VEGF表达与肉瘤恶性度相关 ,相关性可能与VEGF促进血管形成和肉瘤细胞的增殖有关 ,检测VEGF表达可能为判断肉瘤的恶性程度提供帮助。
Objactive:To investigate the relationship between the expression of vascular endothlial growth factor(VEGF) and tumor cell proliferation in sarcoma.Methods:Expressions of VEGF.CD 34 and PCNA proteins in 62 sarcoma with different pathological grade were examined with immunohistochemistry.The expression rate of VEGF,the microvessel density and the grade destribution of PCNA in these sarcomas were made up.ResuIt:The expression rates of VEGF in sarcoma cells with pathological grade Ⅰ?Ⅱ and Ⅲ were 41 2%(7/17)?89 3%(25/28)?94%(16/17),respectively.The microvessel density(MVD)was from 3 to 113.The grade destribution of PCNA expression was:-5 cases(8%),+10 cases(16 1%),++31 cases(50%),+++16 cases(25 8%).The expression of VEGF was positively correlated with pathological grade,MVD and proliferative potential of sarcomas.Conclusions:The detection of expression of vascular endothlial growth factor(VEGF) may help judge the malignancy of sarcoma.
