谷胱甘肽S-转移酶-π在肺癌组织中基因表达和血浆水平的相关性及在肺癌化疗中的指导作用

The correlation between genetic expression of GST-π in the tissue of pulmonary carcinoma and its plasma level and instructive action to chemotherapy in lung cancer

摘要  目的 分析肺癌组织GST-π基因表达和血浆水平的相关性及化疗前后血浆GST-π的水平变化。方法 运用RT-PCR方法对 60例经纤维支气管镜活检肺癌组织及 20例非癌良性肺组织进行GST-πmRNA定量检测,用化学比色法动态测定化疗前后血浆GST-π活力水平。结果 肺癌组织GST-πmRNA表达水平 (1 98±0 76)明显高于良性肺组织表达水平(1 02±0 45),表达阳性率分别为 45%、15%,GST-πmRNA表达与肿瘤组织病理类型有关(P<0 05),组织GST-πmRNA表达(1 98±0 76)与血浆GST-π活力(23 36±7 99)呈正相关(r=0 65,P<0 05),化疗前后血浆GST-π水平均明显高于对照组,化疗前后GST-π水平变化与疗效相关(P<0 05)。结论 肺癌组织恶变过程中GST-πmRNA基因表达及GST-π血浆水平增加,GST-π可以作为肺癌早期诊断的肿瘤标记物;化疗前后GST-π水平变化对临床化疗疗效判断有一定指导意义。 Objective To study the correlation between GST-π expression and plasma level in lung cancer and the plasma level of pre-or post-chemotherapy.Methods The tissue GST-π and the plasma GST-π level of 60 lung cancer patients and 20 noncancer patients were detected by reverse transcription-polymerase chain reaction(RT-PCR)and chemochromatometry respectively.Results GST-π gene expression in lung cancer tissues(1.98±0.76)was higher than that of the noncarcinomatous tissues(1.02±0.45).GST-π gene expression was correlated to pathological type(P<0.05).GST-π gene expression(1.98±0.76)was showed positive correlation with the plasma levels(23.36±7.99)(r=0.65,P<0.05).The plasma GST-π levels of pre-or post-chemotherapy was higher than the control subjects.Correlations were noted between the plasma levels of pre-or post-chemotherapy with therapy effect(P<0.05).Conclusion The genetic expression of GST-π and the plasma levels were increased in the cancerative course of lang cancer.GST-π can be a tumor marker in diagnosing lung cancer.The plasm level pre-or post-chemotherapy has a significant instructive value in estimating the curative effect.
