摘要
目的探讨针对人RON基因跨膜区段(RONm)反义核酸对肺癌细胞系A549活性和细胞增殖的抑制作用,研究以RONm为靶的肺癌基因治疗的可能性?方法将人RONm cDNA反向插入到真核表达载体pcDNA3.1中,转染肺癌细胞A549,利用ELISA检测细胞模型RON基因表达水平,同时利用MTT和细胞计数方法监测细胞生物学活性的变化。结果转染后的肺癌细胞RON基因表达量和细胞活性明显降低,并出现明显的细胞凋亡?结论RONm反义核酸明显抑制RON基因的表达,同时对肺癌细胞的生长有抑制作用,促进细胞凋亡?
Objective: To investigate the effect of In Vitro inhibition of RON gene expression and cell growth effect of lung cancer cell line A549, anti-sense nucleic acid of transmembrane domain of RON gene(RONm) was introduced into this cell line. The possibility of the anti-sense nucleic acid of gene RON as a candidate for lung cancer gene therapy was also explored. Methods: RONm cDNA was inversely inserted in eukaryotic expression vector pcDNA3.1, then the anti-sense nucleic acid of this domain was transferred into the studied cells. The expression of RON gene in this experiment and control groups were monitored by ELISA and the alteration of cell proliferation was checked by MTT assay etc. Results: The expression level of RON gene in the transfected lung cancer cells was reduced and the cell proliferation was inhibited comparing with the control cell lines. Conclusion: The anti-sense nucleic acid of RONm significantly reduces the expression of RON gene in the highly expressed lung cancer cell line A549 and might inhibit the growth of this cell line.
