慢性帕金森病MPTP猴模型自体颈动脉体细胞纹状体移植术后~1H-MRS研究 被引量:1

In Vivo Magnetic Resonance Spectroscopy Study of Chronic Parkinsonian Monkeys by Autotransplants of Carotid Body Cell Aggregates into Striatum

摘要  目的 应用质子磁共振波谱 (1H- MRS) 技术, 进行慢性帕金森病 (PD) 1 甲基 4 苯基 1, 2, 3, 6- 四氢吡啶 (MPTP) 猴模型自体颈动脉体 (CB) 细胞纹状体区移植术后细胞结构生存能力检测。方法 采用 4 只按非人类灵长类帕金森病评分量表 (PPRS) 评价, 行为学症状持续稳定 1 个月的慢性 PD MPTP雄性猴模型, 应用 GE Signa1 5T NV/i磁共振扫描系统进行移植术前定位, 取一侧自体颈动脉体, 使用 Leksell立体导向系统行纹状体区立体定向移植术, 于移植术后 8 周内每 2 周行双侧纹状体区1H MRS 采集, 检测大脑主要代谢物氮 乙酰天门冬氨酸(NAA)、胆碱化合物 (Cho)、肌酸及磷酸肌酸 (Cr), 采用随机软件FuncTool 4 .0对各代谢物含量行半定量分析, 观察NAA/Cr和Cho/Cr的变化, 以配对 t检验行统计学分析, 按PPRS标准进行移植后行为学评分并与代谢物比值进行对照分析。结果 移植术4周后, 移植侧纹状体区 NAA/Cr逐渐增高, Cho/Cr逐渐下降, 术后 6 周与未移植侧纹状体区比较出现显著性差异 (P<0 .05)。行为学评分呈下降趋势。结论 1H MRS能够显示慢性PD MPTP猴模型移植区域的代谢及生化改变, 自体CB细胞移植能够促进局部多巴胺神经元的生长, 可以作为有效手段应用于 PD的治疗。 Objective To evaluate the survival and cellular composition after autotransplants of carotid body cell aggregates into striatum in vivo in MPTP-induced chronic parkinsonian monkeys by using 1H-MRS.Methods Four male MPTP (0.1 mg/kg every day,intracutaneous injection)-induced chronic parkinsoniam monkeys were evaluated by primate Parkinsonism rating scale (PPRS) with stable behaviors for 1 month. Following the localization by MRI scanner,one lateral carotid body cell aggregates were transplanted into single striatum by stereo-localization technique (Leksell,GM592) and the behaviors were evaluated by PPRS 8 weeks after transplantation. After the autotransplant,the 1H-MRS in bilateral striatum were collected with an interval of 2 weeks by GE Signa 1.5T NV/i scanner. The contents of the metabolites including nitrogen-acetylaspartate (NAA),choline-containing compounds (Cho),creatine (Cr) and NAA/Cr and Cho/Cr were semi-quantitatively analyzed by using the FuncTool 4.0 software package. The changes in NAA/Cr ratio and Cho/Cr ratio in the transplant region and unaffected area were analyzed by paired t test and compared with those of behavioral scales. Results Four weeks after transplantation,1H-MRS revealed that the NAA/Cr ratio was gradually increased,while the Cho/Cr ratio gradually decreased in the transplant region as compared with those in the unaffected area. Six weeks after transplantation,the NAA/Cr and Cho/Cr ratio showed significant difference between the transplant region and the unaffected area (P<0.05). The behavioral scales were reduced after transplantation.Conclusion 1H-MRS could noninvasively reveal the biochemical and metabolic alterations in the region with autotransplantation of carotid body cell aggregates in chronic PD monkeys,suggesting that autotransplants of carotid body cell aggregates into striatum could promote the growth of the local dopaminergic neurons and could be used as an effective therapy for PD.
