摘要
目的 研究辛伐他汀对大鼠急性心肌梗死 (AMI)后白细胞介素 (IL ) - 1β基因表达和血流动力学变化的影响。方法 大鼠心肌梗死 (MI)模型分 MI组 (MI- C组 )、MI辛伐他汀干预组 (MI- S组 )和对照组。有创血流动力学检查心脏泵功能 ,逆转录多聚合酶链反应 (RT- PCR)测定非梗死区 IL- 1β m RNA表达 ,Western blot测定非梗死区 IL- 1β蛋白。结果 血流动力学分析显示 ,与对照组相比 ,MI- C组左心室舒张末压 (L VEDP)显著增加(P<0 .0 1) ,收缩压 (SBP)、舒张压 (DBP)、左心室收缩压 (L VSP)和左心室压力最大升降率 (± dp/ dtmax)显著下降 (P<0 .0 5 ) ,而心率 (HR)无变化。与 MI- C组相比 ,MI- S组 L VEDP显著降低 ,± dp/ dtmax明显增加 (P <0 .0 5 ) ,HR,SBP,DBP和 L VSP无差异。 MI- C、MI- S组 IL - 1βm RNA表达较对照组显著增加 (P<0 .0 1) ;MI- S组较 MI- C组明显下降 (P<0 .0 5 )。辛伐他汀显著减少 IL- 1β蛋白质合成。IL- 1β m RNA表达和蛋白质生成与血流动力学的变化呈正相关。结论 辛伐他汀可改善大鼠 MI后心脏血流动力学 ,其机制可能与降低非梗死区 IL- 1β基因表达和蛋白质合成有关。
Objective To investigate the in fluence of simvastatin in indexe s of hemodynamics and expression of IL-1β in rats after acute myocardial infa rction.Methods The myocardial infarction(MI) model of rats were constructed,and div ided into three groups:MI control group (MI-C group),si mvastatin-MI group (MI-S group),sham group.The hemodynamics determined car diac pump function,RT-PCR measured IL-1β mRNA expression in non-infarction reg ion,Western blotting determined IL-1β protein production.Results Compared with sham group,LVEDP significantly enhanced ( P <0.01),SBP,DBP,LVSP and LV pressure ±dp/dtmax markedly lowered respectivel y(P<0.05),but no change of HR in MI-C group.Compare d with MI-C group,LV EDP markedly decreased and LV pressure ±dp/dtmax significantly increased ( P<0.05),while not significant difference of HR,SBP,DB P,and LVSP in MI-S group(P>0.05).The mRNA expressio n of IL-1β markedly in creased in MI-C group compared with sham group(P<0.01 ),and markedly lowered in MI-S group compared with MI-C group ( P<0.05).simvastatin significantly decreased protein production of IL-1β.There existed a positive correlation between mRNA expression of IL-1 β and LVEDP.Conclusion Simvastatin improves hemodyn amics,the mechanism of which may be in part associated with simvastatin decreasing gene expression and protein prod uction of IL-1β in non-infarcted region in post-MI rats.
