摘要
目的 探讨双后肢缺血预处理 (IPC)和肝脏 IPC模型对缺血再灌注 (I/R)损伤肝脏的保护作用及其机制。方法 将 32只家兔随机分为四组 :肝脏 I/R组 (A组 )、双后肢 IPC组 (B组 )、肝脏 IPC组 (C组 )及对照组 (D组 ) ,分别检测其血清丙氨酸氨基转移酶 (AL T)、天门冬酸氨基转移酶 (AST)、碱性磷酸酶 (AL P)、γ-谷氨酰转肽酶 (γ- GT)水平变化 ,以及光镜、电镜下各组肝细胞和细胞器形态学改变 ;并与非 I/R组家兔进行比较。结果 A、B及 C组的 AL T、AST、AL P、γ- GT水平明显高于 D组 (P<0 .0 1,<0 .0 5 ,<0 .0 5 ) ,其中 B、C组明显低于 A组 (P<0 .0 5 ) ,B、C组间无明显差异 (P>0 .0 5 )。B、C组的肝细胞膜及细胞器结构完整 ,A组明显破坏 ,D组均正常。结论 双后肢与肝脏 IPC的肝脏保护作用相似 ,能有效减轻肝脏 I/R损伤。
Objective To investigate the protec tive effect on the ischemic-repe rfusion (I/R) injured liver through the ischemic preconditioning(IPC) on the dou ble hind limbs and the liver,and its mechanism.Methods 32 rabbits were divid ed into 4 groups:liver I/R group (A group),double hind limbs IPC group (B group ),liver IPC group (C group) and normal comparison group (D group),the lev els of alanine aminotransferase (ALT),aspartate aminotransferase (AST),alkalin e phosphatase (ALP) and γ-glutamyl transpeptidase (γ-GT) in serum at every g roup were detected respectively,and the morphological changes in the hepatic cells and hepati c cell devices were observed under the optic and electron microscope. Results The levels of AL T,AST,ALP and γ-GT in group A,B and C were significantly higher than those in group D(P<0.01,<0.05,<0.05).There was no signi ficant difference betwe en group B and C,P>0.05.The structures of the hepati c cells and hepatic cel l devices were intact in B and C,were significantly destroyed in group A,wer e normal in group D.Conclusion The IPC on the double h ind limbs has the same protective effect for the I/R injured liver as the IPC on the liver and can effectively decrease the I/R injuries on the liver.
