重复手术法收集超数排卵后的家兔卵母细胞 被引量:6

摘要 采用促卵泡素(FSH)和FSH混合25%聚乙烯吡咯烷酮(PVP)两种方法进行超数排卵,以正常排卵母兔作对照,经过3次重复剖腹手术(每次间隔至少50天以上)收集家兔卵母细胞,旨在探索大量获取家兔成熟卵母细胞简单有效的方法。结果超数排卵的两组获取总卵母细胞数和可用卵数比对照组均高出3倍以上(P=0.001),两种超数排卵效果无显著差异(P>0.1)。3次连续手术均成活的家兔中,对照组和FSH组3次取卵效果无显著差异(P>0.05),FSH混合25%PVP组首次手术与第三次手术卵母细胞数出现显著差异(P<0.01)。本方法可有效获取大量家兔卵母细胞,并提高实验动物利用率。 Rabbits were superovulated by conventional six injections (3d) of follicle-stimulating hor mone (FSH) or by a single subcutaneous injection of FSH dissolved in a 25%solution of polyvinylpyrrolidone(PVP),and no-superovulate females were control group.The oocytes were recovered from both superovulated and no-treated females by laparotomy.Rabbits were given the same gonadotrophin treatment and 1aparotomy up to three times at intervals of at least 50 days.The results showed that the two superovulatory groups resulted in over 3 times of the overall number of recovered oocytes when compared with the control group(P=0.001),and with no signifcant differences in two superovulatory groups(P>0.1).Among the living rabbits after 3 consecutive times surgery,the efficiency of recovery in FSH treatment group and control protocols did not differ(P>0.05).However,at the FSH-PVP treatment group,themean numbers in the third surgery was significantly lower than the first(P<0.01).So it indicated that the methodology used in the present study could be an efficient way for obtaining 1arge numbers of high quality oocytes in rabbit and improved the usage rate of laboratory animals.
