摘要
为阐明肝硬化时TXA_2/PGI_2的代谢状况及其与门脉高压的关系,我们研究了慢性胆管结扎犬肝硬化时血浆TXB_2/6-keto-PGF_(1α)变化及其与门脉血液动力学的关系。结果表明,肝硬化犬下腔静脉TXB_2水平与正常犬相近(P>0.05),肝静脉显著高于正常犬(P<O.001),门静脉显著低于正常犬(P<0.01);肝硬化犬下腔静脉、肝静脉和门静脉内6-keto-PGF_(1α)水平均显著高于正常犬(P<O.001),而TXB_2/6-keto-PGF_(1α)比值则显著低于正常犬(P<O.001)。正常大血浆TXB_2/6-Keto-PGF(1α)水平及其比值与门脉血液动力学无相关性。肝硬化犬下腔静脉6-keto-PGF_(1α)水平与Q_(TL)呈显著负相关(r=-0.74,P<0.05)、TXB_2/6-keto-PGF_(1α)比值与Q_(HA)呈显著正相关(r=0.85,P<0.01);肝静脉TXB_2(r=0.81,P<0.05)和TXB_2/6-keto-PGF(1α)(r=0.85,P<0.01)与Q_(HA)呈显著正相关;门静脉内TXB_2、6-keto-PGF(1α)水平及其比值与门脉血液动力学无相关性。结果表明,肝硬化犬有明显TXA_2、PGI_2代谢异常,TXA_2在维持肝硬化时肝动脉有效血液灌注中可能起有益作用。
To explore the metabolic state of A_2/PGI_2 and their relations to portal
hypertension in cirrhosis, splanchnic plasma levels of TXA_2 and PGI_2 were measured by
RIA in chronic bile-duct-ligation-induced cirrhotic dogs. The results showed that, in the
cirrhotics, TXA_2 content of inferior vana cava was not elevated, but that of hepatic vein
was significantly increased, and that of portal vein was notably decreased as compared with
the controls. Prostacyclin concentrations of inferior vana cava, hepatic and portal veins in
the cirrhotic dogs were much higher than that of the control dogs, but the ratio of TXA_2
to PGI_2 was notably reduced compare with the normal. There was no relation found between
TXA_2 or PGI_2 and portal hemodynamics in the control. However, a negative correla-
tion between PGI_2 and total hepatic blood flow and a positive correlation between TXA_2
or TXA_2/PGI_2 ratio and hepatic arterial blood flow were found in the cirrhotics. These
results suggested that the cirrhotic dogs have an abnormal TXA_2/PGI_2 metabolite and
TXA_2 may play a beneficial role in the maintenance of efficient hepatic perfusion in cir-
rhosis.
