摘要
目的探讨雄激素对成骨细胞代谢的影响。方法胎鼠头盖骨成骨细胞培养于含不同浓度睾酮的培养基中,观察细胞胸腺嘧啶核苷掺入、碱性磷酸酶(ALP)活性、骨钙素基因表达及骨钙素合成等细胞增殖及分化等指标的变化情况。结果胎鼠头盖骨成骨细胞的增殖不受睾酮影响,但ALP活性、骨钙素基因表达及其合成在不同程度上受睾酮的调节。结论雄激素对成骨细胞的分化具有促进作用。
Objective\ To investigate the effect of testosterone on bone cell metabolism.Methods\ Fetal rat calvarial osteoblastic cells were cultured in the medium containing different concentrations of testosterone. 3H thymidine incorporation,alkaline phosphatase(ALP) activity,osteocalcin gene expression and production were detected to determine the condition of cell proliferation and differentiation. Results \ The proliferation of fetal rat calvarial osteoblastic cells was independent of the presence of testosterone.However,ALP activity,osteocalcin gene expression and production were regulated in different extent by testosterone. Conclusion\ Testosterone stimulates the differentiation of rat calvarial osteoblastic cells in vitro .
