摘要
为探索IL-6(Interlmkin-6)是否在IDDM(insulin-depenednt diabetes mellitus.胰岛素依赖的糖尿病)的发病中起作用,本文报告了我们采用反转录多聚酶链反应(RT-PCR)对IDDM 病人,NIDDM(Non-insulim dependent diabetes Mellitus,非胰岛素依赖的糖尿病)病人,正常儿童(Normal chidren,NC)正常成人(Normal adults,NA)四组受试者的外周血单个核细胞(peripheral blood mononuclear cells,PBMNCs)中的IL-6 mRNA 的表达水平初步研究。以内源性β_2微球蛋白(β_2-microglobulim,β_2m)的mRNA 作为内对照,与靶序列IL-6 mRNA 在同一管内同时反转录及扩增,以特异的PCR 产物产量之比IL-6/β_2m 表示IL-6 mRNA 表达的相对水平,结果显示:在12例IDDM 病人,29例NIDDM 病人,23例正常儿童及12例正常成人的PBMNC_a中,IL-6 mRNA 的相对水平分别为0.91±0.19;0.10±0.06;0.43±0.08,0.01±0.07。这个结果提示IL-6很可能在IDDM 的发病过程中扮演重要角色。
Whether or not an abormal expression of IL-6 mRNA in PBMNCs from IDDM patientswas examined using a hihgly sensitive,specific and semiquantitative protocal,i.e.reverse tran-scription and polymerase chain reaction (RT-PCR).The relative levels of IL-6mRNA in PBM-NCs from 12 early IDDMpatients (8.20±3.85yr),29 newly diagnosed NIDDM patients(54.85±9.12yr)23 normal childrens (8.20±3.26yr) and 12 normal adults (31.92±11.22yr)weredetermined.Significantly high expresion levels of IL-6 mRNA were found in PBMNCs from pa-tients with IDDM (P<0.05,T-test).The reletive levels of mRNA in each group were 0.91±0.19,0.10±0.06,0.43±0.08,0.10±0.07,respectively.Our data suggest that higher expres-sion of IL-6 mRNA in PBMNCs from patients with IDDM may reflect that IL-6 plays an impor-tant role in the pathogenesis of IDDM.
