摘要
动脉内皮细胞(endothelial cells,EC)损伤在动脉粥样硬化(AS)的发生,发展中极为重要。血液中胆固醇浓度增高,特别是低密度脂蛋白(low densitylipoprotein,LDL)含量增加可引起动脉EC损伤,导致前列腺环素(prostacyclin,PGI_2)合成障碍,从而与血栓素A_2(TXA_2)平衡失调,促进AS的发生发展。本文的研究目的是评价丹参酮ⅡA(tanshinone ⅡA,TN ⅡA)对LDL引起的EC损伤的影响。
It has been demonstrated that low density lipoprotein(LDL)may damage endothelial cells(ECs)resulting in atherosclerosis.The purpose of this study was to evaluate the effect of tanshinone Ⅱ A(TN Ⅱ A)on LDL induced EC damage.ECs was randomly divided into 4 experimental groups(n=8)and treated with medium 1640 alone,2mg·ml-1 LDL.2mg·ml-1 LDL plus 1μmol·L-1 TN Ⅱ A,2 mg·ml-1 LDL plus 100μmol·L-TN Ⅱ A,respectively.Then they were incubated with LDL or TN Ⅱ A for 36 h.At the end of the incubation period,the media were collected to analyze the contents of PGl2 TXA2 and malonyldialdehyde(MDA)in ECs.It was observed that MDA and TXB2 were increased greatly and 6 keto PGF1α was decreased in the LDLgroup.TN Ⅱ A enhanced the 6 keto PGF1α level and suppressed the rise of the MDA content.The ratio of TXB2/6 keto PGF1α was regulated by TN ⅡA.The results confirm that LDL may increase LPO,resulting in EC damage and suggest that TN Ⅱ A could protect ECs by preventing the cytotoxic LPO accumulation in EC membranes produced by LDL.
