摘要
用于抗癌活性评价的MTT法采用“SDS-异丁醇-HCl”溶解液后可完全溶解甲(月朁),也可完全溶解细胞阳血清蛋白等成份,得到澄明、稳定的甲(月朁)溶液(A_(570)值稳定时间不少于12h)。本法测得的抗癌活性量-效关系十分显著,与同位素核酸前体参入法结果吻合。本法用于悬浮细胞的药敏测定时更为方便、可靠。
'SDS-isobutanol-HCl' dissolving solution was used in the MTT assay. This 'triplex solution' could not only dissolve formazan fully but also dissolve cells and serum proteins completely, giving ■ clear and stable formazan solution (A_(570) values were consistent within 12h). The concentration-response relationships were marked and consistent with the results obtained by the isotope labeling precursor incorporation assay. This improved MTT method was more convenient and reliable, especially when being used in suspended cell survival determination.
