摘要
本文通过高压液相技术对矽肺大鼠肺泡巨噬细胞上清中的促纤维母细胞生长活性物质进行了初步分离纯化。结果表明,SiO_2诱发的矽肺大鼠肺泡巨噬细胞上清中,出现一异常增高的蛋白洗脱峰,分子量范围在6-9KD之间,能明显促进同糸大鼠肺纤维母细胞的生长,放射免疫分析表明,这一洗脱峰具有较强的胰岛素样生长因子-1(IGF-Ⅰ)的免疫反应性。其次,对矽肺大鼠支气管肺泡灌洗液及血清的分析表明,灌洗液中IGF-Ⅰ升高明显,但血清IGF-Ⅰ水平无变化。
In present study we demonstrate that the promoting fibroblast growth activity in the supernatant of silicotic
rat alveolar macrophages is attributed to insulin—like growth factor—Ⅰ mainly.Partially purification of super-
natants of alveolarmacrophages,which were from silica exposed in vivo five to six—week rats.revealed a protein
peak eluted from molecular—sieve HPLC column.peak 5.corresponding to M.W.around 6——9 KD.Activity
assay and radioimmunoassay indicated this peak is more potent to stimulate fibroblast growth and have higher
insulin—like growth factor—Ⅰ immunoresponsibility.but no any detectable activities of interleukin—1 and tumor
necrosis factor.Quantification of insulin—like growth factor—Ⅰ level in silicotic rat bronchoalveolar lavage fluids
and tended to increase as the prolongation of silica exposed in vivo,but no alteration of insulin—like growth fac-
tor—Ⅰ level be found in sera.
