摘要
原生质体的分离与培养是食用菌原生质体融合工作的重要基础。本文比较了不同溶壁酶组成及浓度、渗透压稳定剂、pH值、反应温度、菌丝培养基成分和菌丝的生理状况等因素对香菇(Lentinus edodes)菌丝原生质体分离的影响。用PDY培养基培养的幼嫩菌丝,在1.0%溶壁酶和1.0%脱壁酶的混合酶液系统中,以0.6M KC1 为渗透压稳定剂,pH5.0,反应4小时可形成10~7/ml以上的原生质体。在再生培养基中加入香菇菌丝提取液可获得较高的原生质体再生率。
<Abstract > Isolation and regeneration of protoplast is a fundamental basis of protoplast fusion in edible fungi. The effects of different osmotic stabilizer, pH, temperature, enzymes, concentration, culture media and mycelium age on the formation of mycelium protoplast as well as regeneration of Lentinus edodes were studied. The highest protoplast yield was obtained by using PDY medium, combined enzyme system containing 1.0% Lywallzyme and 1.0% Lyric enzyme. 0.6M KCl, pH 5.0 and reacting 4 hours. The result also showed that the extract of mycelium added to the regeneration medium can increase the ratio of proto- plast regeneration.
