乳腺癌导致机体乙酰化率升高机制的初步探讨

摘要 本文应用高压液相色谱(HPLC)法,测定血样、肝匀浆上清液及酶反应产物中对氨基苯甲酸(PABA)的乙酰化率。观察结果:①移植乳腺癌MA782/5S瘤株后12天,小鼠血中乙酰化率较基础值平均增加了7.93%(n=20)(P<0.001),同时,肝乙酰化率较对照组平均增高20.05%(n=10)(P<0.001);另外单纯测定一批荷瘤小鼠的肝乙酰化率也较对照组平均增高21.64%(n=10)(P<0.001)。②体外实验中,6次测得人乳腺癌BCaP─37细胞内乙酰化酶活性Vm为每分钟0.98±0.036nmol/mg蛋白。以上结果提示荷瘤小鼠乙酰化率升高的原因可能为:①肿瘤组织本身具有乙酰化代谢能力;②小鼠荷瘤时肝脏乙酰化能力也会增强。 A High-pressure Liquid chromatography (HPLC) method has beenused for the detection of acetylation rate of PABA in blood samples, hepatic cell cytosoland enzyme-reaction products in this study. The resuLts showed that:1)the acetylation rate in blood was increased by 7.93% on average(n=20) 12 daysafter inoculation of carcinoma MA782/5S in Kun-ming mice (p<0.001), at the same time,the liver acetylation rate was higher by 20.05 % than that in the control group (n=10) (p <0.001).In addition, the liver acetylation rate was determined in an other batch of tumor-bearing mice and control mice the result showed that the liver acetylation rate was also higher by 21.64%(n=10) than in the control group (p<0.001).(2) In vitro,the mean acetyltransferase activity of human mammary tumor BCaP-37cells obtained from six seperated experiments was 0.98 ± 0.033nmol/ min/mg. The reasons for the increased acetylation rate in tumor-bearing mice may be:(1) tumortissuse has the ability of acetylation ;(2) acetylation ability of the liver increases at thetumor─bearing status.
