摘要
本文介绍用醋酸纤维素-戊二醛-己二胺-抗体抗原膜与专用原电极研制的lgM非标记免疫传感器.采用均匀设计法优化了膜结合抗原抗体的实验条件,研究了抗体浓度对电极灵敏度及线性范围的影响,并对抗原抗体结合时间、测试介质等条件进行了选择.与同类电极相比,该电极使用方便、响应快、灵敏度高、重现性和稳定性较好.在最佳测试条件下,电极线性范围47~850μg/mL,相关系数0.9973.用于血清样品测量,取得满意结果.
This thesis constructed a nonlabelled IgM Immunosensor by combining anacetylcelluose-glutaraldehyde-hexanediamine-antibody-antigen membrane with a specialoriginal electrode. The homogeneous design was used to optimize experimental condi-tions of bounding antibodies (antigens) to membranes. Effects of antibody concentra-tions on electrode sensitivity were studied, the time which antigen-antibody reactionneeds nad determinate mediums were also selected. Compared with other immunoelec-trodes, this one is convenient to operate and fast to respond. Its sensitivity, repro-ducibility and stability are perfect. On the optimum conditions, the linear rang of thiselectrode is 47-850 μg/mL,the correlation cofficient is 0. 997 3. Satisfactory results wereobtained by using this electrode to determine IgM in human sera.
