摘要
本文介绍了两种简便、快速的DNA提取方法,它们不需要特殊试剂、液氮和柱层析系统、超速离心机等贵重仪器。我们以海刀豆和红树两种盐生植物为实验材料,所制得的DNA样品经紫外分光光度计检测:其最大吸收波长均为258nm;A_(258)/A_(280)均为1.82;与Sepharose4B柱层析分离结果相近;用琼脂糖凝胶电泳检测为均一的谱带。
Two simple, quick methods of DNA extraction are introduced here. Special reagentand valuable instruments, such as liquid nitrogen, column chromatography and ultracentrifugeetc. were not required in the extraction. Two kinds of salt-enduring plants-Canavalimamaritima and Rhizophora apiculata were chosen as experimental materials. DNA samplesobtained were checked by ultraviolet spectrophotometer and agrose electrophoresis. Both of theirmaximum absorption wavelength were the same of 258 nm; Both Of their A_(258)/A_(280) ratio were 1.82; the isolation results closed to that by Sepharose 4B. The electrophoresis gel tests showed thatthere was an equal and even one band.
