高血糖与醛糖还原酶的激活 被引量:1

摘要 应用荧光法、C18反相高压液相层析和气相色谱法分别测定17例正常人和32例Ⅱ型糖尿病患者(17例有并发症)红细胞内的醛糖还原酶活性、某些核普酸和糖醇浓度。结果显示:Ⅱ型糖尿病患者红细胞内醛糖还原酶的活性高于正常人;有并发症患者的酶活性又高于无并发症患者,证明高血糖能激活醛糖还原酶,激活程度与血糖浓度呈正相关。患者红细胞内NADP+、山梨醇和果糖的浓度高于正常对照;NADPH和NAD+浓度低于正常对照,而此5项指标在两组患者间差异无显著性。 In this paper we determined the activity of aldose reductase,and the concentration ofsome nucloetides,sugars and polyols in the erythrocytes of 17 controls and 32 patients withType II diabetes mellitus(NIDDM)by the fluorometric method,C18 reverse phase HPLC andgas chromatography respectively.Among the 32 patients,15 cases had no complications,theremainders had various complications,The activity of aldose reductase in NIDDM patientswas significantly higher than that of controls(P<0.05 and P<0,01 respectively)and thatof patients with complications was significantly higher than those without complications(P<0.05).Hyperglycemia can activate aldose reductase and positive correlation existed be-tween sugar concentration and aldose reductase activation(r=0.90,P<0.01).The concen-trations of NADP+,sortitol and fructose in the erythrocytes of NIDDM patients were signifi-cantly higher than those of controls(P<0.01,P<0.05 and P<0.05 respectively).The con-centrations of NADPH and NAD+in the controls were significantly higher than those of pa-tients(P<0.01 and P<0.05),while there was no difference between these two groups ofpatients P>0.05).The NAD+ decrease is perhaps due to insulin-resistance of cells in NID-DM patients,therefore causing decline of glucose oxidation.
