摘要
将鸡马立克氏病毒(MDV)血清2型Z_4毒株第10代(Z_4-10),在对鸡马立克氏病(MD)高度易感的P系SPF鸡胚成纤维细胞(CEF)单层上连续传代全第30代(Z_4-30),Z_4毒株空斑形成速度和空斑形态均未发生显著变化。分别以Z_4-11,Z_4-20,Z_4-25和Z_4-30细胞结合毒作单价疫苗,将Z_4-11,Z_4-20和Z_4-30细胞结合毒分别与F_(C126)细胞结合毒组成二价疫苗,免疫P系SPF鸡,用MDV强毒GA株攻击,进行免疫效力试验。结果表明,Z_4毒株4个代次单价苗之间以及3个代次毒分别与F_(C126)毒株组成的二价疫苗之间免疫效力无显著差异。表明Z_4毒株在CEF单层上从第10代连续传至第30代,仍然保持其原有的培养特性和免疫原性。
Different passages of Z_4 strain in chicken embryo fibrablast(CEF)culture were tested to evalu-ated the stability of characteristics in culture and immunogenicity of the virus Z_4 strain of serotype 2 MDV,Z_4strain was serially passaged from passage 11 to passage 30 in CEF nomolayer cuIture originated form line P(B19/B19) SPF chickens which are highly susceptible to Marek’s disease(MD).The time interval requiredfor developing plagues, the average size and morphology of plaques, in defined period of time post-inocula-tion did not change siginificantly for Z_4 strain up to passage30.Protective efficiencies of four Z_4 monovalentvaccine, F_(C126) monovalent vaccine and bivalent vaccines respectively containing passage 11 of Z_4 and F_(C126), pas-sage 20 of Z_4 and F_(C126) or passage 30 of Z_4 and F_(C126) were compared, and the results showed that immunogenic-ity of Z_4 strain remained unchange up to passage 30.
