摘要 从裂褶菌SchizolhyllimcommuneFr.菌株的菌丝体,提取水溶性多糖、醇析、Sevag法脱蛋白、透析、再径SephadexA—50,SephadexG—200柱层析纯化,得裂褶多糖Spg(1).用酸水解、部分酸水解、酶解、PC、TLC、GC、IR、H-NMR、NaIO4氧化,Smith降解等分析,证明Spg是一种β─葡聚糖。主链β(1→3)连接,支链β(1→6)连接,分子量10万. The Schizophyllum commune Fro of produced Schizopyllan have been obtained through tissue isolation from field fruit bodes of Schizophyllum commune Fr. which was collected from the rot wood of broasleaf trees in Zijin mountain, Naming. The mycelia of S. commune of a 2 week- old agar slant culture on PDA medium were inoculated to 100 ml of soybean meal glucose liquid medium in a 500 ml shaking flask and the flask incubated on a rotary shaker at 120r/ min at 26°C for 3 d to produced the preculture. The same type of flasks with the 1000 ml of liquid medium were then inoculated with 50 ml of preculture and incubated for 4 d at 26 C, the culture broth of all flasks was transferred to a fermentahon tank with 3500 L of liquid medium and the fermentation period was 3 d at 26 C. The fermentation broth is extremely viscous, with fruit aromatic. It was separated into mycelial cake and culture filtrate through centrifuge.The crude Schizophyllan was prepared by ethanol precipitation from the hot water extract of submerged cultrue mycelia of Schizophyllum commune Fr. The proteins of crude Schizophyllan were removed by the Sevag procedure, the isolation and purification of Schizophyllan through DEAE-Sephadex A-50 and Sephadex G-200 column chromatography; an endocellular pure Schizophyllan designated as Spg was obtainbed.The chemical structure of Spg has been determinated by techniques of physical and chemical analysis. The infrared spectrometry analyse show a characteristic absorption peak in 890 cm-1 and the 'H-NMR chemical shifts i&less than 5 ×10-6. The analytical results above proved that Spg possess β-glucosidic linkages. From the results of complete of partial hydrolysis and enzymolysis, periodate oxidation, smith degradation, Spg has been shown to be aglucan consistng of β-1.3- and β-1.6-glucosidic linkages. The molecular weight was estimated by gel filtration to be 100000.The Spg is whit6 fibrous polysaccharide to be easily soluble in Cold or hot water forming an extremely viscous solution, [α] = 12.5, pH 6-7, element analysis: C 42.6, H 6.64.
