摘要
采用新型Ca ̄(2+)荧光指示剂Fura-2建立双波长荧光法测定分离的大鼠神经细胞内游离钙浓度([Ca ̄(2+)]_i).结果显示,在静息状态下,其[Ca ̄(2+)]_i为109±12nmol/L.30mmol/LKCl可显著增加[Ca ̄(2+)]_i,并且KCl的这种效应呈一定的剂量依赖关系,提示该法灵敏、可靠.
he intracellular free Ca ̄(2+) concentration([Ca ̄(2+)]) was measured in isolated neurocytesprepared from Sprague-Dawley rats with Fu-ra-2 double-wavelength fluoremetry. The re-sults showed that the [Ca ̄(2+)]i, of the restingneurocytes in Ca ̄(2+)-containing solution was 109+12nmol/L(-x±s, n=9).KCl (30mmol/L)markedly evoked[Ca ̄(2+)]of the neurocytes(P<0. 01, n=9),and the rises of [Ca ̄(2+)]i byKCl is a concentration-dependent response.These results suggest that the adoption of Fu-ra-2 double-wavelength fluoremetry in dissoci-ated rat neurocytes in a useful and relativelyeasily applicable technique for monitoring in-tracellular Ca ̄(2+) changes.
