摘要
进行高效液相色谱 -质谱联用测定人血清中地红霉胺浓度的方法学及其药动学研究。血清样品经0 1mol·L-1氢氧化钠溶液 0 .1ml碱化 ,乙酸乙酯提取 ,采用HypersilODS2色谱柱 ;乙腈∶甲醇∶水 (6 2∶30∶8,V V V)流动相 ;流速为 1.0ml·min-1。质谱条件为AP -ESI离子源 ,正离子模式 ,电喷雾电压 35psig ,干燥气 (N2 )流速 10.0L·min-1,干燥气温度 32 0℃ ,毛细管电压 4 0 0 0V ,碎片电压 170V ,选择性监测质荷比 (M Z)为 735.5的分子离子峰 (M +H)。结果 :在 5.0~ 12 0 0 . 0ng·ml-1范围内地红霉胺血清浓度呈线性关系 ,方法回收率大于 90 %。试验制剂地红霉素肠溶胶囊相对生物利用度F为 97.0 3%± 9.98%。高效液相色谱 -质谱联用测定人血清中地红霉胺浓度 ,灵敏、准确 。
A HPLC-MS method was established to determine the concentration of erythromycylamine in human serum. A Hypersil ODS2 column was used with acetonitrile-methanol-water (62∶30∶8). The flow rate was 1 0ml·min -1 . AP-ESI and M/Z of 735 were selected to quantify erythromycylamine. The linear range was 5~1200ng·ml -1 and the determination limit was 0 5ng·ml -1 .The method was sensitive, fast and accurate, it could be used in the monitoring of therapeutic drugs and human pharmocokinetic study of dirithromycin.
