摘要
研究了影响多胺测定的苯甲酰化反应的时间和温度,以及用反相高效液相色谱法测定柑橘愈伤组织多胺的条件.结果表明,苯甲酰化反应的最佳条件为 37 ℃,反应 30 min.采用 Waters C18反向柱(4.6 mm×250 mm,5 μm),G1314A 紫外检测器,以甲醇-水溶液(体积比为 64∶36)为流动相,流速 0.7 mL/min,柱温 30 ℃,检测波长 230 nm,外标法定量.该法灵敏、准确.
This paper is an attempt to study the reacting time and temperature of benzoylation affecting the detection of polyamines,and the detecting method for polyamines in Citrus callus with reversed phase high performance liquid chromatography.Reacting 30 min at 37 ℃ were the optimal benzoylation conditions.Chromatographic separation was performed in Waters C18 column(4.6×250 mm,5 μm) with the thermostat at 30 ℃,using a methanol-water(64∶36, volume ratio) mixture as mobile phase with a flow rate of 0.7mL/min,and the eluate was monitored with a G1314A UV detector at 230 nm.Quantities were calculated with external standards.The method was found to be sensitive and accurate.
