建立了测定对羟基苯海因(HPHT)的反相高效液相色谱法。固定相为 Spherigel ODSC18柱,流动相为乙睛 水 四氢呋喃 冰乙酸,体积比为 70∶30∶05∶0.5,检测波长为 254nm。方法的线性范围为0.1~17.5 mg,回归方程 A=2 062.3 m-293.3,相关系数为 0.999 5,相对标准偏差为0.86%,检出限为4.3μg,加标回收率为99.3%~102.0%。
RP-HPLC was applied to the determination of 5-(4-hydroxyphenyl)-hydantoin (HPHT) and the following chromatographic conditions were optimized. Spherigel ODS C_(18) column (4.6 mm×250 mm, 5 μm) was used as the stationary phase. A mixture of CH_3CN, H_2O, THF and HOAc in the ratio (by volume) of 70∶30∶0.5∶0.5 was used as the mobile phase. UV-detection was made at the wavelength of 254 nm. Linear relationship was obtained between the peak area and the mass of 5-(4-hydroxyphenyl)-hydantoin in the range of 0.1 to (17.5 mg), giving the following regression equation: A=2 062.3 m-293.3 (r=0.999 5). The detection limit was found to be 4.3 μg. RSD (n=10) found at the concentration level of 1.0 g·L^(-1) was 0.86%, and the recoveries tested by the standard additions method were in the range from 99.3% to 102.0%.
