难溶性药物盐酸尼卡地平单室单层渗透泵片的制备 被引量:10

摘要 目的 制备难溶性药物盐酸尼卡地平的单室单层渗透泵片 ,体外维持 2 4h恒速释放。方法 以高、低分子量聚氧乙烯、氯化钠组成片芯 ,以CA为包衣材料 ,制备盐酸尼卡地平 (NP)单室单层渗透泵片 ,并对片芯组成进行优化。结果 片芯最佳组成为 :NP 80mg ,PEO(Mr 2 0 0 0 0 0 ) 5 6mg ,PEO(Mr 5 0 0 0 0 0 0 ) 4 2mg ,氯化钠 10 0mg。本渗透泵片 2 4h的累积释放率 95 % ,并在 2 4h内维持零级释放。结论 本渗透泵片主药、辅料比例适宜 ,制备简便 ,能较好地控制难溶性药物释放达 2 OBJECTIVE: To prepare the monolithic osmotic pump tablet (OPT) of nicardipine hydrochloride. METHODS: Monolithic OPT of NP was prepared using low or high moleculan weight PEO and NaCl as core, CA as the coating material. The core formulation was optimized. RESULTS: The optimal core formulation consisted of NP 80 mg, PEO(Mr 200 000) 56 mg, PEO(Mr 5 000 000) 42 mg and NaCl 100 mg. The drug was released from OPT at controlled rate 24 h, and the cumulative release percentage 24 h was 95%. CONCLUSION: It provided the new and practical technology for controlled drug release for 24 h.
