摘要
目的 研究姜黄素对急性粒细胞白血病HL 6 0细胞体内、体外抗癌作用。方法 应用台盼蓝拒染法、MTT法及电镜观察超微结构和DNA片断化分析观察姜黄素对HL 6 0细胞的体外抗癌作用 ;通过HL 6 0细胞裸鼠移植瘤模型 ,采用TUNEL ,免疫组化等方法检测姜黄素对HL 6 0细胞裸鼠移植瘤的体内抗癌效应。结果 ①姜黄素对HL 6 0细胞的增殖具有明显的抑制作用 ,呈时间和剂量依赖性 ;在体外可诱导HL 6 0细胞发生凋亡。②姜黄素抑制HL 6 0细胞裸鼠移植瘤生长 ,抑瘤率为 5 4 35 % ;姜黄素在体内亦可诱导HL 6 0细胞凋亡 ,并使其Bcl 2蛋白的表达水平下降。结论 姜黄素在体内、外均可抑制HL 6 0细胞增殖 ,并诱导其凋亡 ;可能通过下调Bcl 2蛋白表达起作用。
OBJECTIVE: To study the anticancer activities of curcumin in HL-60 cells in vitro and in vivo. METHODS: The effect of curcumin on the proliferation and apoptosis of HL-60 cells in vitro was studied by trypan blue dye exclusion, MTT assay, ultrastructure and DNA fragmentation analysis. The anticancer effect of curcumin on xenograft growth of HL-60 cells was evaluated by tumor inhibition rate, histology, TUNEL and immunocytochemistry. RESULTS: 1Curcumin inhibited the proliferation of HL-60 cells in vitro in a time dependent and dose dependent manner. Curcumin induced HL-60 cells apoptosis that was confirmed by DNA fragmentation and ultrastruction. 2Curcumin inhibited xenograft growth of HL-60 cells with the inhibition rate of 54.35%. There were more apoptic cells in curcumin treatment group than that in the control group. The expression of Bcl-2 protein was down-regulated in the curcumin treatment group compared with the control group. CONCLUSION: Curcumin was able to inhibit the proliferation of HL-60 cells and induce HL-60 cells apoptosis in vitro and in vivo by down-regulating the expressin of Bcl-2 protein.
